SUMMARY:

The trypsin nhibitor isolated from tamarind seeds (TTI) has been explored for potential applications in the treatment of non-communicable chronic diseases (NCDs). However, toxicological safety still needs to be investigated to ensure the proposed applications. Therefore, this study examined the toxicological safety of TTI by administering two concentrations at different stages of zebrafish life. Initially, TTI was obtained through protein fractionation by ammonium sulfate. Fraction 2, which showed higher inhibitory activity against trypsin in the range of 30-60% (F2), was isolated by Trypsin-Sepharose 4B Affinity Chromatography. Fish in both the embryonic and adult stages were exposed for 96 hours to three concentrations of TTI (12.5, 25, and 50 mg/L). Various analyses were conducted to identify acute toxicity and assess cardiotoxicity, hepatotoxicity, neurotoxicity, and genotoxicity. Embryos exposed to TTI were investigated for survival, hatching, malformations, and motor visual response in 7 days post-fertilization (dpf) larvae. Adult fish underwent behavioral tests after acute exposure for 96 hours; however, the experimental tests are still ongoing. TTI showed no acute toxicity in zebrafish embryos, and both concentrations improved the motor visual response in 7dpf larvae, suggesting a potential protective effect of TTI, with additional results pending. Therefore, the study provides a thorough assessment of the safety of TTI, a crucial step in ensuring its potential applicability in human health.
