SUMMARY:

Molecules of plant origin have been studied for their antibacterial activities and among them are trypsin inhibitors. Soon the need arose to carry out a systematic review on mechanisms of antibacterial action of trypsin inhibitors and to evaluate the in silico interaction by molecular dynamics of purified trypsin inhibitor from tamarind seeds (TTIp) with bacterial membranes. In order to carry out the review, a protocol for obtaining a systematic review was initially prepared and registered on the International Prospective Platform for Systematic Reviews (PROSPERO) and an article related to the protocol was produced, aiming at guiding the execution of the systematic review, available in the registry: CRD42020189069. This systematic review with meta-analysis will be developed in accordance with the Preferred Report items for Systematic Reviews and Meta-analysis (PRISMA-P). Original articles resulting from studies with rats and / or mice, in vitro (bacterial culture) and cell studies, describing treatment and the antibacterial mechanisms of peptides or proteins of the type inhibitors of will be included. trypsin. Searches will be carried out in the following databases: PubMed; ScienceDirect; Scopus; Web of Science, EMBASE and the Virtual Health Library (VHL) Regional Portal and the selected articles were evaluated using the Ryyan tool. The risk of bias will be assessed using the SYRCLE tool (Systematic Review Center for Experimentation with Laboratory Animals). Methodological quality in animals will be assessed using the CAMARADES checklist (Collaborative Approach to Meta-Analysis and Review of Animal Data from Experimental Studies). The quality of the studies and the risk of cell bias will be assessed by the OHTA (Office of Health Assessment and Translation). If at least two studies show clinical and / or methodological and / or statistical homogeneity, a meta-analysis will be performed, using the RevMan Analyzes statistical package in Review Manager v.5.3. It is expected to find several manuscripts highlighting the mechanisms related to the action of peptides or proteins inhibiting trypsin on infectious diseases caused by bacteria. In addition, an in silico study of the interaction between ITTp and bacterial membranes using the lipid bilayer model using the CHARMM-GUI server, using molecular dynamics using the Gromacs 5.1.2 package is being conducted. The knowledge of these mechanisms will precede the development of new research and also, the foundation for the creation of innovative products.
