RESUMO:

Nanoencapsulated carotenoids appear as an alternative to reduce the inflammatory processes triggered by the high consumption of ultra-processed foods. Thus, the objective of this study was to evaluate the safety and bioactivity of nanoparticles based on a crude carotenoid-rich extract of Cantaloupe melon and porcine gelatin in Wistar rats. Encapsulation was performed by oil/water emulsification followed by freeze-drying, and porcine gelatin was used as an encapsulating agent and Tween 20 as a surfactant. Different physical and chemical analyses were done to characterized powder particles (EGS) and were evaluated the incorporation efficiency (EI). Toxicity was investigated by signs of general toxicity and food intake in male Wistar rats (n = 5 per group) overfed with a high glycemic index (HGLI) diet. And subjected to the following conditions during 10 days of study: dietary supercharged HGLI and treated with 12.5 mg crude carotenoid extract (EB) / kg animal weight in water by gavage; overfed with a HGLI diet and treated with 50 mg nanoencapsulated carotenoid (EGS) / kg animal weight in water by gavage. Besides, to the control groups without overfed treatment with HGLI diet and treated with a standard diet. Blood toxicity (hemogram, lipid profile, liver, and renal function), liver and small intestine morphology and histopathology were evaluated. EGS characterization analyzes showed the presence of spherical particles with a smooth surface, mean diameter of 74,00 (12,70) nm, polydispersion index of 0,58 (0,14), and EI of 98% (1.78). As for signs of general toxicity, there was no significant difference (p> 0.05) between the initial and final weight of the rats of each group evaluated. For food consumption, a significant increase (p <0.05) was observed only for the obese group without treatment. Regarding blood toxicity, it was verified by the biochemical parameters analyzed low EGS toxicity. The morphological and histopathological analyses of the liver showed acute hepatitis in the animals of all groups evaluated, and those treated with EB and EGS presented tissue repair. The results obtained for the absolute and relative liver weight determinations showed that there was no significant difference (p> 0.05) between the studied groups. For small intestine, chronic enteritis was observed in all animals, with considerable improvement in villus and intestinal gland structure in animals treated with EGS. Besides, the absolute weight evaluation indicated a significant reduction in the intestine of the animals in the untreated group [6.58 (0.39)] (p <0.05) compared to the others evaluated. The results suggest the safe application of EGS and bioactive effect, considering the results observed for the intestine. This evidence possibly linked to the anti-inflammatory potential, showing the safety regarding its use in future clinical studies, aiming to investigate the efficacy in controlling mucosal-related intestinal inflammatory processes.
