RESUMO:

Allium cepa L. and S-methylcysteine have been investigated because of the positive effects on diabetes mellitus attributed to antioxidant and anti-inflammatory functions. The objective of the study was to evaluate the antioxidant properties and the modulation of the inflammatory response of Allium cepa L. and S-methylcysteine in rats with experimental diabetes. Thirty-five Wistar rats with streptozotocin-induced diabetes mellitus (50mg / kg i.p.) were used. The groups were divided into: CT: non-diabetic animals; DM: animals with untreated experimental diabetes; DMAC: animals with experimental diabetes treated with Allium cepa L .; DMSM: animals with experimental diabetes treated with S-methylcysteine. Allium cepa L. extract (400mg / kg body weight) and S-methylcysteine (200mg / kg body weight) were administered daily for 30 days. The analyzed variables included weight, polydipsia and polyphagia; metabolic parameters, pancreatic and hepatic morphology and morphometry; antioxidant activity and markers of inflammatory response. The results were expressed as Mean (Standard Error). Among the treated groups, differences were observed only for triglycerides (TGL) and VLDL, p<0,05. The DMAC and DMSM groups had the lowest values of TG: 97.5 (20.0) mg / dL and 79.2 (19.1) mg / dL, p <.05, respectively; and VLDL 12.3 (4.0) mg / dL and 15.8 (3.6) mg / dL, p <0.05, respectively, when compared with DM 170.0 (20.0) and 32 , 4 (5.4) mg / dL. Mean values of fasting glycemia were lower (524.5 (9.3) mg / dL, p <0.05) in the DMSM group, compared to DB (700.4 (13.9) mg / dL). Among the analyzed cytokines IL-1, IL-6, IL-10, TNFα, an increase in the concentration of IL-10 (1.004.3 (169.3) pg / ml, p <0.05) was observed only in the DMSM group, when In the morphological analysis, there was a change in hepatic glycogen deposition in the DMAC (3.9%, p <0.01) and DMSM (4.1%, p <0.05) groups compared to the DM group (393.7 (21.2) pg / The DMAC and DMSM groups recovered the atrophy of the pancreatic islets observed in the DM group. The activity of superoxide dismutase was increased in the DMAC groups (327.3 (56.4%)). ), And the DMSM (409.9 (77.8) UA / min / mg prot, p <0.01), compared to the DM group (109.2 (AU / min / mg prot, p <0.05) The same was observed with catalase, which was elevated in the DMAC and DMSM groups (4,957.8 (777.9) nmol / min / mg prot, p <0.01 and 5.111, The results showed that the treatment with Allium cepa extracts was associated with an increase in the number of treatments, L. demonstrated have positive effects on tissue morphology, lipid profile and antioxidant activity. Treatment with S-methylcysteine was more effective in promoted an improvement in glycemia and modulation of the inflammatory response. These results suggest that the compounds have beneficial functionalities in the treatment of diabetes mellitus.
