RESUMO:

Research to combat obesity has focused on the use of diets similar to the dietary pattern of the current population to induce disorders in animals. However, no studies in Wistar rats have consistently associated the use of high carbohydrate content pellet-diets to dysfunctions in the adipose tissue and intestinal epithelium, linking inflammation in different important tissues to obesity.  This study investigated the effect of a high glycemic index and glycemic load (HGLI) pellet-diet on intestinal and adipocyte morphology, its presence in liver and pancreas and its relation with inflammation in Wistar rats. Two groups of adult animals (n = 5) received for 17 weeks: (1) HGLI Diet or (2) Standard Diet (Labina®). Microscopic analysis of adipose tissue, central portion of the small intestine, liver, and pancreas was performed. Stereology, gene expression, immunohistochemistry, and TNF-α dosage were analyzed. The HGLI diet induced not only hypertrophy of adipocytes (1387 μm) but also inflammation and changes in the small intestine.  Fat deposition in the liver and pancreas, a 5.9-fold greater expression of TNF-α in adipose tissue, positive immunostaining (+2) and high serum TNF-α (6.4 μg / ml) was also observed in rats under the HGLI diet. Thus, this diet was able to generate remodeling in the adipocytes with fat deposition in different organs, inflammation, and a disorder in the intestinal villi. These features are commonly demonstrated in the use of fat pellet-diets or high fructose-rich beverages. Therefore, we conclude the HGLI diet is an excellent instrument for the use in research aiming to induce the alterations demonstrated here in Wistar rats.
