RESUMO:

Obesity is a worldwide epidemic that can be caused by genetic, environmental, and metabolic / hormonal factors. Among the hormonal factors, it is seen as the individual in a state of obesity, presented as a response to an evaluation of certain hormones in relation to eutrophic individuals, such as: hyperleptinemia, leptin resistance, as well as the change in the performance of the Cholecystokinin (CCK). Because it is a multifactorial disease, its treatment also occurs by several means, among which the use of CCK secretagogues stands out among several possibilities. This study aimed to evaluate the nutritional status and effect of ITT as CCK secretagogue and its synergism with leptin in obese Wistar rats. An experimental study with three groups of obese Wistar rats (n = 15) was carried out, using the standard commercial and high glycemic index (HGI Diet) and ITT diets for 10 days. . Obese rat groups were administered: HGI diet plus water (hyperglycemic diet + 1 mL water per gavage); HGI plus ITT (hyperglycemic diet + 1 mL of ITT per gavage at the concentration of 25 mg / kg) and standard diet plus water (standard diet + 1 mL of water per gavage) and verified during that period the effect of ITT on Nutritional status of obesity, serum level and expression of CCK-related genes at the intestinal and leptin level in adipose (subcutaneous and visceral) tissue. It was observed that all animals were obese before and after the experiment, however, mice that underwent ITT treatment had a tendency to reduce Lee's index when compared to the untreated group and standard treatment. Obese animals treated with ITT had a significant increase in CCK1R receptor expression, however, no difference was observed between gene expression values for CCK (Pre-pro-CCK), CCK2R receptor and convertase enzyme ( PSCK1). However, there was a significant decrease in CCK1R receptor expression for animals treated with the standard diet that interestingly did not differ from that treated with ITT. It was also observed that the animals treated with ITT presented lower serum leptin values than those treated with a nutritionally adequate diet and the HGI diet, with a statistical difference in the ob gene expression level. Therefore, in the study of the effect of ITT on serum leptin levels and results related to the secretagogue effect of ITT to CCK with obese animals, a synergistic action is suggested in this study. In that ITT by reducing leptin levels, it improves the action of CCK in obese animals, resulting in the tendency to reduce the nutritional status of obesity demonstrated in this study.
