Quantification of genes involved in immune response to gastrointestinal nematodes in sheep

Gastrointestinal nematodes infections take to losses in animal production, especially in sheep farming, an important socioeconomic activity. Undestanding the mechanisms involved in the immune response of these animals to these infections is essential, because animals belonging to the same breed and herd respond differently to infections. The lack of studies involving this differentiation in sheep leads to the need to better undestand these mechanisms. In this sense, the aim of this work was to quantify the expression of genes involved in the immune response (IFN-γ, TNF-α, IL-4, IL-13, IL-9, IL-22, TGF-β e IL-10) to gastrointestinal nematodes in sheep phenotypically classified as resistant and susceptible. Abomasum, small and large intestine samples from 12 crossbred sheep ½ Santa Inês x ½ without a defined breed pattern, naturally infected by gastrointestinal nematodes, 6 resistant group (RG) and 6 susceptible group (SG) animals were used. From these samples, total RNA was isolated from the tissues, synthesized cDNA, which was quantified by qPCR to determine relative expression, using na ideal reference gene for each tissue (GAPDH, SDHA and RPL-19) and analyzed by REST© software. In abomasum, IL-4 (2,5x) and IL-13 (1,1x) were upregulated in RG and IFN-γ (1,2x), TNF-α (1,1x), IL-9 (2,6x), IL-22 (2,8x), TGF-β (3,5x) (P<0,006) e IL-10 (1,4x) were up regulated SG. In small intestine there IFN-γ (3,4x), IL-4 (2,8x) (P<0,017), IL-13 (1,3x), IL-22 (1,6x) were up regulated in RG and TNF-α (5,3x) (P<0,001), IL-9 (1,8x), TGF-β (1,0x) and IL-10 (1,1x) in SG. In large intestine, IFN-γ (2,3x) (P<0,030), IL-4 (1,3x), IL-13 (7,0x) (P<0,0001), IL-22 (3,0x), TGF-β (2,6x) (P<0,001) and IL-10 (2,8x) (P<0,003) were up regulated in RG and TNF-α (2,2x) e IL-9 (1,5x) in SG. In conclusion, except for the Th2 response which was observed in all tissues of the resistant group and IL-9 which was up regulated in the tissues of susceptible group, there was no predominance of a specific immune response in the different tissues due to parasite dynimics in each organ be different.
