SUMMARY:

The resistance to antifungals has been increasing in some groups of fungi
that cause invasive mycoses. Typically, the selection of resistance occurs through prior treatment with
antifungals. However, there are reports of infections caused by already resistant fungi, without previous
exposure to the antifungal. This raises the hypothesis that the process of acquiring resistance may occur in
the environment, possibly selected by a drug with antifungal effects, such as environmental fungicides and
hospital disinfectants. The objective of this study was to investigate the resistance profile to drugs with
antifungal activity in medical importance yeasts found infecting/colonizing patients, in animals, and in the
environment, testing the hypothesis of cross-resistance between clinical and environmental antifungals.
Yeasts were isolated from different sources and identified by mass spectrometry (MALDI-TOF MS). A total of
135 yeasts were isolated and identified, with a prevalence of species from the genus Candida (101; 74.8%),
Kodamaea (17; 12.6%), Cryptococcus (7; 5.2%), and Trichosporon (5; 3.6%). After identification, theusing the broth microdilution method with the clinical antifungals fluconazole, amphotericin B, and
micafungin. The yeasts of the genus Candida exhibited 9.6% resistance to clinical antifungals. The highest
resistance was attributed to micafungin (6.9%) and occurred in Candida glabrata. Three strains showing
multidrug resistance were also found, resistant to both amphotericin B and micafungin. In the next stages of
the project, cross-resistance analysis and sequencing of resistant strains will be conducted. It is expected
that this study will provide crucial data on antifungal resistance in yeasts, helping to address whether this is
a problem that must be faced from a One Health perspective, aiding in the development of more effective
prevention and treatment strategies.
