SUMMARY:
Supramolecular complexes are organized systems that result from the spontaneous association of two or more chemical species held together by intermolecular forces. Inspired by the same principle, this project proposes the development and characterization of nanoparticles based on chitosan (CS) and carboxymethyl-β-cyclodextrin (CM-β-CD). CS is a cationic polyelectrolyte widely used in biomedical applications which is able to interact and form supramolecular complexes with oppositely charged molecules as CM-β-CD, anionic modified cyclodextrin. These systems are capable of improving the pharmacokinetics of both hydrophilic and lipophilic drugs. CM-β-CD was synthesized from β-CD and characterized by infrared spectroscopy (FTIR), thermal analysis (TGA), X-ray diffractometry (XRD) and degree of substitution determined by mass spectroscopy (MALDI -TOF MS). The nanoparticles dispersions of CS and CM-β-CD were characterized in terms of viscosity, particle size (light scattering), conductivity and potential ζ; and the physicochemical characterization was performed by the FTIR, XRD, TGA and scanning electron microscopy (SEM). The nanoparticles with resveratrol (RES) were characterized in terms of FTIR, XRD, TGA/DSC, DLS and potential ζ; and an isothermal titration calorimetry (ITC) was used to evaluate the thermodynamic aspects of the interaction, such as: stoichiometry, entropy and enthalpy changes and constant affinity between cyclodextrins and drug, and between chitosan and cyclodextrins.
