Several studies report that memory and synaptic plasticity are facilitated in genetically modified mice expressing a constitutively active form of the α1A adrenoceptor subtype (α1A-AR). However, there is a paucity of research employing pharmacological tools to investigate the role of this adrenoceptor type in cognitive processes, possibly due to the lack of pharmacokinetic evidence to help understand the distribution of α1A-AR ligands across tissues, including passage through the blood-brain barrier (BBB). The aim of this study was, therefore, to evaluate the effect of the α1A-AR agonists, oxymetazoline and cirazoline, on recognition memory consolidation in mice. Initially, an in silico analysis was performed to predict the permeability of the compounds across the BBB. Subsequently, male Swiss mice received acute oral treatment with vehicle, cirazoline (0.0001, 0.001, 0.01, 0.1, and 1 mg/kg), or oxymetazoline (0.01, 0.1, and 1 mg/kg) immediately after the Object Recognition Test (ORT) training session to assess memory. In silico analysis predicts that both compounds have a favorable profile for crossing the BBB. In behavioral assays, treatment with oxymetazoline, under the conditions tested, did not promote improvement in recognition memory. In contrast, cirazoline, at doses of 0.01 and 0.1 mg/kg, significantly increased exploration of the novel object compared to the familiar one, providing a robust pro-mnemonic effect. Neither treatment altered the animals' total exploratory activity, ruling out interference from motor effects. It is concluded that pharmacological activation of the α1A-adrenoceptor, specifically by cirazolin, represents an effective strategy for improving the consolidation of long-term recognition memory.
