SUMMARY:

The use of plants for medicinal purposes has been practiced since ancient times through the use of various plant parts. It is used as a complementary therapeutic source. Geoffroea decorticans, popularly known as Chañar, is a plant native to Chile belonging to the Fabaceae family. Its bark, flowers, fruit, and leaves have uses in folk medicine with known expectorant activity, including bronchopulmonary disorders and pain relief, as well as antioxidant and antinociceptive activities with the fruit. However, few studies are reporting such activities with extracts of Chañar seed. That said, our work aimed to evaluate and characterize the antioxidant, cytotoxic, and protective potential against oxidative stress induced by copper sulfate (CuSO4) of the ethanolic (EE) and aqueous (EA) extracts of Chañar seed using in vitro and in vivo methodologies. Firstly, a phytochemical characterization (HPLC - GC-MS/MS) was carried out on these two extracts, as well as spectrophotometric tests such as total phenolic compounds and total flavonoids. Subsequently, biochemical tests were carried out to evaluate the in vitro antioxidant potential utilizing five spectrophotometric tests (CAT; Reducing power; DPPH; Copper and Iron) which aimed to verify the possible modes of action by which these bioactive compounds could be contributing to the maintenance of cellular redox homeostasis. The NHI-3T3 fibroblast cell line was also used to evaluate the cytotoxicity of EE and EA, as well as the protective role of the extracts when this cell line was subjected to the stressor copper sulfate (CuSO4), in addition to the potential to induce cell migration by observing the closure of a wound. The extracts were also tested on an invertebrate animal model (Tenebrio molitor). Survival tests were carried out to assess the toxicity of the extracts and their protective capacity using the copper sulfate oxidative stress test, as well as assessing melanization and insect development parameters. The results obtained by chromatography detected secondary metabolites such as phytol, alpha-tocopherol, vitexin and rutin. The in vitro spectrophotometric tests demonstrated the antioxidant capacity of the extracts to act by different mechanisms of action (inhibiting, sequestering and chelating). Meanwhile, tests on the NIH/3T3 cell model revealed the absence of EE and EA cytotoxicity, as well as protection against (CuSo4) stress. In the animal model, the extracts once again showed their excellent protective capacity against oxidative stress, capable of fighting free radicals generated, mitigating the melanization exacerbated by stress and the lack of modulation in the insect's metamorphosis process.  Taken together, the tests (in vitro and in vivo) may indicate that the EE and EA extracts of Chañar seeds could be a possible source of bioactive compounds with excellent antioxidant properties. This would also strengthen the use of the by-products generated by the plant, thus boosting its consumption, given that depending on the consumption of the fruit, the seeds are considered a waste product of the crop. In addition to providing scientific evidence for the user population, governed by cytotoxic phytochemical studies.
