Diabetes mellitus (DM) is a chronic metabolic disorder characterized by persistent hyperglycemia, resulting from deficiency in insulin production by the pancreas and/or its action, leading to long-term complications. Pharmacological therapy for DM faces limitations due to the high number of side effects. Alternative treatments based on natural products have potential in the treatment of DM. The species Spondias tuberosa Arruda, known as umbuzeiro, has been widely used in traditional medicine to treat digestive diseases, infections and diabetes. Despite its frequent use, there is little scientific exploration of its benefits. However, a recent study demonstrated its antihyperglycemic activity. Considering the easy access to this species in the region and its pharmacological potential, the objective of this work is to evaluate the antidiabetogenic effect of Spondias tuberosa Arruda. Clinical, biochemical, morphological analyzes and evaluation of possible markers involved in the therapeutic effect were carried out in diabetic rats treated with this species. Fifty-two Wistar rats were used, and diabetes mellitus was induced by streptozotocin (40mg/kg i.p.). The groups were divided into 5: GC (non-diabetic animals), DM (animals with experimental diabetes), EX (non-diabetic animals treated with S. tuberosa extract), DMEX (animals with experimental diabetes treated with S. tuberosa extract) and DMIN (animals with experimental diabetes treated with insulin). S. tuberosa extract (500 mg/kg, p.o.) and NPH insulin (10UI s.c.) were administered daily for 30 days after the onset of diabetes mellitus. The parameters analyzed in this study include clinical evaluation where there was an increase in water intake, food consumption and urinary volume, with weight loss in animals induced by STZ in relation to the control group, biochemical profile where there was a significant decrease in the hepatic enzyme AST of the group treated with the extract in relation to the group without treatment, morphology and morphometry of the pancreas where the animals treated with the extract showed greater similarity with the animals in the control group in relation to the group without treatment, and oxidative evaluation of the liver and immunomodulatory activity of the pancreas that is yet to be performed. It was found that STZ-induced DM caused polyphagia, polydipsia and polyuria; STZ destroyed pancreatic beta cells, causing atrophy and changes in the morphology of pancreatic islets as well as increased positive staining for PAS in the liver; Treatment with S. tuberosa partially recovered the morphology of pancreatic islets and improved liver biochemical parameters (ALT) as well as having similar results to GC in PAS marking; Insulin treatment was able to normalize hyperglycemia, polyphagia, polyuria, polydipsia and partially improve the morphology of pancreatic islets. Investigation of this plant will contribute to a better understanding of the complex pathophysiology of DM1 and will encourage research into new therapeutic or complementary plant-based alternatives medicinal products from the rich Brazilian flora.
