RESUMO:

Diabetes Mellitus is a worldwide epidemic that is expected to increase in the coming decades. It is a heterogeneous metabolic disorder and is classified as three major forms: DM1, DM2 and gestational diabetes. The experimental DM1 is the focus of this study. DM1 is characterized as an autoimmune disease that causes the immune-mediated destruction of pancreatic β cells, resulting in insulin deficiency, leading to hyperglycemia which in turn leads to damages such as retinopathy, neuropathy, gastrointestinal disorders, among others. The objective of this study is to evaluate the resulting alterations from DM1 in the duodenal portion and the possible protective effects of the extract of Allium cepa L. and its isolated amino acid S-methylcysteine in its morphology. For the purpose of this study, 35 male Wistar animals, aged 90 days and weighing between 250 and 300 g, were used. The animals were allocated in polypropylene boxes with drinking fountain and feeder. They were divided into four groups: CONT (control), DMST (diabetic group without treatment), DMSM (diabetic group treated with Smethylcysteine) and DMAC (diabetic group treated with Allium cepa L extract). The treatment period lasted 30 days, then the animals were euthanized with a lethal dose of isoflurane for sample removal. The samples were fixed for routine histological processing with HE staining to determine volume using the Cavalieri principle, proportion occupied by the duodenum layers using the Delesse principle, absolute volume and immunohistochemical staining for IL-10, TNF- α, Caspase 3 and BCL-2. The treatments reduced the glycemia in the DMSM, DMAC groups compared to the DMST group, but had no protective effect on weight loss, polydipsia and polyphagia. As to the increase in mucosal volume and total volume seen in diabetic animals, the treatments reduced those parameters, being more effective on the group treated with the extract. From the results, it was concluded that the treatments were able to reduce glycemia and partially reverse the morphometric alterations when comparing treated and untreated diabetic animals. Thereby, it can be concluded that phytotherapic intervention can act as a coadjuvant alternative in the treatment of diabetes with both pre-clinical and duodenal tissue level benefits.
