RESUMO:

Diabetes Mellitus (DM) is a metabolic disease characterized by hyperglycemia, which leads to the increase of reactive oxygen species (ROS) and, consequently, to oxidative stress. These changes lead to morphofunctional complications in different organ systems, including the heart, in which diabetic heart disease (CD) is identified. For the reduction of oxidative stress, treatment with antioxidant agents that could act in the protection of the organism in diseases like DM have been used. The aim of this study was to analyze the morphofunctional changes in the ventricles of rats due to DM and the antioxidant treatment of quercetin (100mg / kg), glutamine (1%) and vitamin E (1%). Twenty male Wistar rats with 150 days of age were divided into four groups (n = 5): N (normoglycemic without treatment); D (untreated diabetic); NS (normoglycemic supplemented with antioxidants) and; DS (diabetic supplemented with antioxidants). The results demonstrated that DM promoted reduction of final body weight in D and DS. There was an increase in cardiac weight / body weight ratio, water intake, feed intake and glycemia in D and DS. Blood levels of IL-1β and TNF-α were similar between groups, but in relation to IL-10 there was reduction in D and increase in DS. The volume density (Vv) in the heart of collagen fibers was higher in D and DS, but lower in the supplemented group. FGF-2 + was increased in D and DS. There was higher expression of SOD-1 + in D and DS, whereas GPX + was higher in D and lower in DS. There was an increase in the expression of IL-1β + and TGF-β + in D and DS when compared to C, although a slight reduction of these in DS was observed. The data suggest that antioxidant supplementation minimizes body mass loss and decreases cardiac hypertrophy but does not contribute to improved polydipsia, hyperphagia, and hyperglycemia. At the systemic level, DM promoted a decrease in the activation of anti-inflammatory cytokines, while antioxidant supplementation increased their activity. Increased expression of fibroblasts and decreased collagen deposition in DS suggesting a remodeling process compared to D. At the tissue level there was an increase in oxidative stress (expression of SOD-1 +) in D and DS, and a decrease in the markers antioxidants in DS (expression of GPx +). In relation to the inflammation markers, there was an increase in these in the diabetic groups, but a slight reduction in the DS group compared to D was observed, suggesting that supplementation with quercetin, glutamine and vitamin E initiates the process of reducing local inflammation even before decrease of oxidative stress.
