ASSOCIATION OF CYP2R1 AND VDR POLYMORPHISMS WITH METABOLIC SYNDROME-RELATED COMPONENTS IN BRAZILIAN ADOLESCENTS

SUMMARY:

Associations between vitamin D deficiency, Metabolic Syndrome and Metabolic Syndrome-related componets have been demonstrated by epidemiological studies, however the biological mechanisms remain controversial. The aim of this study was to investigate the association between variants of the CYP2R1 and VDR genes and metabolic syndrome-related components in Brazilian adolescents. This cross-sectional study included 174 adolescents classified as overweight. Three CYP2R1 gene variants and four VDR gene variants were measured by allelic discrimination. The distribution of different genotypes and alleles were compared between different groups. It was observed that the GG genotype and the G allele of the rs12794714 polymorphism (CYP2R1 gene) were significantly associated with an increased risk of Metabolic Syndrome. The GG genotype of the rs10741657 polymorphism (CYP2R1 gene) was significantly associated with increased risk of hyperglycemia (OR = 3.89, 95% CI 1.17-12.91, p= 0.026). The AG+GG genotype of the rs2060793 polymorphism (CYP2R1 gene) was also associated with hyperglycemia. Furthermore, the CC genotype of the rs7975232 polymorphism (VDR gene) was significantly associated with an almost 6-fold increase in the risk of hypertension (OR = 5.91, 95% CI 1.91-18.32, p= 0.002). In conclusion, there was an increase in the risk of MS and risk of hyperglycemia in adolescents who had polymorphisms of the CYP2R1 gene. Furthermore, we observed that, the rs7975232 polymorphism of the VDR gene was significantly associated with the risk of hypertension.
