SUMMARY:

Green propolis, a widely consumed natural substance, has demonstrated an effect on the bone in several ways. To evaluate the effect of alcoholic green propolis extract on orthodontic tooth movement in rats. The experiment will follow the second guidelines of Animal Research Reporting In Vivo Experiments – ARRIVE. In this randomized in vivo experimental study, 40 healthy male Wistar rats (Rattus norvegicus) will be used, between 7-10 weeks, 150-250g. They will undergo orthodontic tooth movement through a closed nickel-titanium helical spring activated between the left 1st molar and upper incisors and daily administration of the substance by gavage for 21 days. The animals will be distributed into four groups (n = 10 in each): control group (GA): orthodontic tooth movement and daily dose of water; experimental group 1 (P1) – orthodontic tooth movement and daily dose of green propolis (100mg/kg); experimental group 2 (P2) – orthodontic tooth movement and daily dose of green propolis (300mg/kg); experimental group 3 (P3) – orthodontic tooth movement and daily dose of green propolis (500mg/kg). The amount of orthodontic tooth movement and bone volume will be analyzed using microcomputed tomography (Micro-CT). Scores will be assigned as (0) absent; (1) level; (2) moderate; and (3) intense to analyse inflammatory infiltrate by hematoxylin and eosin (HE) staining. For root resorption, tartrate-resistant acid phosphatase (TRAP) staining will be used – osteoclast count will be performed. Bone remodeling will be proven by Osteoprotegerin (OPG), RANK and RANK L immmunoexpression. Statistical tests will be carried out at a significance level of 5%.
