SUMMARY:

Although in-office tooth bleaching with highly concentrated hydrogen peroxide is effective, sensitivity and immediate mineral loss are still factors to overcome. This study aims to develop and characterize nanostructured desensitizers loaded with pharmacological active ingredients associated with a remineralizing agent and evaluate the transdental permeability in vitro. The desensitizers will be prepared using the phase inversion method, with droplet size ≤ 170 nm and negative zeta potential. Physical stability tests, loading tests, rheological and characterization properties will be carried out. To evaluate the transdental pharmacological permeability, dental fragments will be made from the proximal surfaces of human third molars, divided into seven (n=7). Group 1 (control - without active ingredient); Group 2 (drug 1); Group 3 (drug 2); Group 4 (drug 1 + drug 2); Group 5 (drug 1 + remineralizing agent); Group 6 (drug 2 + remineralizing agent); Group 7 (drug 1 + drug 2 + remineralizing agent). The fragments will be fixed in artificial pulp chambers, whose inner surface of the dentin will be in contact with distilled water. The nanogels will be applied before and after bleaching (5 minutes) with 35% hydrogen peroxide (3 applications of 15 minutes). The transdental permeability of the drugs will be evaluated by reading the concentration of active ingredients in distilled water using the High-Performance Liquid Chromatography (HPLC) technique one hour after the final removal of the nanogels.
