SUMMARY:

The renin-angiotensin system is of great importance for the body's water balance and blood pressure regulation, in addition to being associated with the stimulation of pro-inflammatory pathways. Its main peptide is angiotensin II, which interacts mainly with type 1 (AT1) and type 2 (AT2) receptors. This study aimed to evaluate molecular aspects in mice submitted to an experimental model of periodontal disease, observing the influence of Ang II type 1 (AT1) and Ang II type 2 (AT2) receptors on periodontitis. Methods: Experimental periodontitis was placed by placing a ligature with 5.0 nylon thread around the upper left second molar of AT1, AT2 and wild-type (WT) mice, subdivided into 2 groups for each strain: no ligature and ligature, totaling six groups: three controls and three experimental. After 15 days of disease induction, the animals were euthanized. In order to assess whether genetic variations had an influence on periodontal disease, analyzes of cytokines, peptides and enzymes were performed from gingival tissues by ELISA and RT-PCR. Results: IL-1β levels significantly increased in the WT and AT2 linked groups compared to the NL (p < 0.01 and p < 0.05, respectively), while in the AT1-L group the levels decreased when compared to WT-L (p < 0.05). For IL-6 there was an increase in the WT-L and AT2-L groups compared to controls (p < 0.001), while the levels for AT1-L increased (p < 0.001). IL-10 levels were higher in the WT-L group (p < 0.01) compared to the control group, WT-NL. For TNF-α, the wild-type and AT2 groups showed increased levels (p < 0.001 and p < 0.01, respectively), while AT1 showed a reduction (p < 0.001). For Angiotensin II and Angiotensin 1-7, the group with the highest levels was AT2-L compared to its control (p < 0.01 and p < 0.01, respectively). All groups with periodontitis had an increase in the relative expression of TLR2 RNA (WT: p < 0.05, AT1-NL: p < 0.01 and AT2-NL: p < 0.01), whereas for the receptor But the the WT-L and AT2-L groups showed an increase (p < 0.001 and p < 0.001, respectively). For ACE expression was higher in the WT-L group (p < 0.001).
