SUMMARY:

The altered expression of tumor suppressor genes and cell cycle regulatory proteins are related to the etiopathogenesis of different cancers. Tumor suppressor genes encode proteins that reduce the probability of a cell to become neoplastic, among them, there is the family of growth inhibitors (ING), which inhibit uncontrolled cell growth. Cell cycle regulatory proteins act on its progression, resulting in cell proliferation; cyclin D1 protein, for example, represents an important regulator of this cycle. Oral epidermoid carcinoma (OEC) is a malignant neoplasm of complex pathogenesis, while oral epithelial dysplasia (OED) is an entity that, depending on its evolution, exhibits risks of malignant transformation for OEC. The aim of this study is to compare the immunoexpression of ING3 and cyclin D1 proteins between the biological behavior of OEC and OED. In all cases, gradation will be performed according to the criteria suggested by the World Health Organization (WHO), in addition to the gradation of Kujan et al. (2006), for OED, and the gradation of Almagunsh et al. (2015) for OEC. The immunohistochemical analysis will be performed quantitatively, considering the cellular location (cytoplasmic and/or nuclear) of the proteins studied. For the statistical analysis, associations will be made between the immunoexpression of Cyclin D1 and ING3 comparing the two groups of samples, with clinicopathological variables through Fisher’s Exact, Kruskal-Wallis (KW) and Mann-Whitney (U) statistical tests. For OEC cases, overall (OS) and disease-free (DFS) survival rates will be evaluated using the Kaplan-Meier method and compared by the log-rank test. A significance level of 5% will be adopted for all tests considering the value of p ≤ 0.05. With the development of this study, we seek to understand the role of ING3 and Cyclin D1 proteins in oral carcinogenesis.
