SUMMARY:

Oral tongue squamous cell carcinoma (OTSCC) is a malignant neoplasm that develops from the lining epithelium and shows poor prognosis, low survival rates, rapid local invasion, and a propensity to early lymph node metastases. Despite the great progress in recent decades, the understanding of the biological and molecular processes involved in this lesion is not completely elucidated. In this way, researchers continue to search for molecular biomarkers with predictive value to identify high-risk patients, as well as to enable the development of new therapeutic strategies. The Hippo pathway has been extensively studied because of its functions in the control of organ size, homeostasis, and tissue regeneration, as well as in the carcinogenesis process when there is inactivation of this pathway. There are several proteins involved in the activation and inactivation of this pathway. In mammals, the main proteins MST1/2 and LAST1/2 show as main functions the regulation of the transcription coactivators YAP and TAZ. Depending on the phosphorylation status of the YAP/TAZ complex, this pathway can be directed towards a tumor suppressor or a pro-oncogenic effect. Thus, the Hippo pathway proteins have been studied as possible markers of tumor progression, prognosis, and therapeutic targets in some neoplasms since they are involved in the regulation of several cellular processes. Therefore, this research aims to evaluate the immunoexpression of LATS2 and YAP1 in cases of OTSCC, correlating their expressions with clinicopathological parameters and patient survival. The sample will consist of 60 cases of OTSCC and 20 cases of normal oral mucosa. To assess the morphology of OTSCC cases, the depth of invasion (DOI) incorporated into the T parameter of the TNM staging system will be used, as well as the gradations systems proposed by WHO (2005) and by Almangush et al. (2014). The immunohistochemical profile of LATS2 and YAP1 will be evaluated by scores based on their intracellular location (cytoplasm and/or nucleus) and epithelial distribution. For the analysis of the studied parameters, the Pearson's Chi-square, Fisher's exact, Mann-Whitney, and Kruskal-Wallis statistical tests will be performed. Survival analysis will be performed using the Kaplan Meier estimator and the log-rank test. For all evaluations, values of p < 0.05 will be considered significant.
