SUMMARY:

Odontogenic lesions, represented by odontogenic cysts and tumors, present a high percentage among those that affect the oral cavity, and some exhibit a more aggressive clinical-biological behavior, being locally invasive and exhibiting high recurrence rates. Invasiveness and recurrence are some of the mechanisms associated with the epithelial-mesenchymal transition (EMT), a process in which epithelial cells gain phenotypic properties characteristic of mesenchymal cells, including greater motility and invasiveness. Thus, through regulation by EMT-associated transcription factors, such as Snail / Slug, Twist, and Zeb, a reduced expression of epithelial proteins associated with overexpression of mesenchymal proteins is evidenced. Thus, the present work aims to analyze the immunohistochemical expression of proteins involved in the EMT process (Zeb1, E-cadherin, N-cadherin, and vimentin) and to correlate them with clinical and pathological aspects involved in specimens of odontogenic cysts and tumors. The sample object of the study will be represented by 90 cases of lesions of odontogenic origin, consisting of 30 cases of ameloblastoma, 30 of odontogenic keratocyst, and 30 of dentigerous cyst. The collection of clinical information such as age, sex, location, size of the lesion, and recurrences will be carried out. All specimens submitted to immunohistochemical processing will be analyzed through scanned slides (Aperio CS2 Digital Pathology Scanner-Leica Biosystems), with a random choice of 5 (five) fields, which will be photographed at 200x magnification (Case Viewer). The expression of each marker, based on its analysis in its specific cell compartment, will be evaluated by multiplying the scores related to the staining intensity (0 = negative, 1 = weak, 2 = moderate and 3 = strong) by the scores associated with the percentage of immunolabeled cells (1 = 1-25%; 2 = 26-50%; 3 = 51-75%; and 4 = ≥76% of positive cells). Based on the findings observed in this study, we intend to better understand the pathogenesis of the evaluated lesions and establish a possible relationship between MET and their biological behavior.
