SUMMARY:

During oral carcinogenesis, malignant cells acquire an aggressive phenotype that results in increased individual motility and the ability to invade surrounding tissues. Therefore, malignant epithelial cells develop a regulatory and programmed process called epithelium-mesenchymal transition (EMT), which is crucial for the acquisition of this aggressive malignant phenotype. During this process, epithelial cells lose part of their characteristics and acquire others inherent to mesenchymal cells, a process orchestrated by several transcription factors inducing these changes. The aim of the present study is to investigate the role of immunohistochemical expression of EMT signaling proteins in oral epithelial dysplasias and oral tongue squamous cell carcinomas, as well as to identify possible associations of the regulation of the Twist gene with the expression of EMT biomarkers and the ability to invasion, migration, proliferation and cell viability in vitro. For the immunohistochemical study, cases of epithelial dysplasias and oral squamous cell carcinomas will be selected, in which the immunoexpression of the proteins Twist, Snail, E-cadherin and N-cadherin will be analyzed. Possible associations between the pattern of expression of these proteins with the histopathological gradation of epithelial dysplasias and with the different histopathological degrees, clinical aspects and survival in oral squamous cell carcinomas will be investigated. For the in vitro study, cell cultivation of two cell lines of oral squamous cell carcinoma, SCC-25 and HSC-3, will be carried out, in which the regulation of the Twist gene will be made, evaluating possible associations with the expression of the biomarkers of EMT Snail, E- cadherin and N-cadherin using Western Blotting and qRT-PCR techniques, as well as analyzing the ability of migration and invasion through functional tests. With the results obtained in this study, we intend to contribute to a better understanding of events that signal malignant transformation in lesions with this potential in oral language, in addition to advancing on the knowledge of the role of EMT proteins as possible biomarkers of stages of carcinogenesis and of progression in oral squamous cell carcinoma.
