The oral cavity harbors a complex microbiome, generally in dynamic equilibrium, the disruption of which can favor the growth of pathogenic microorganisms, leading to periodontal diseases. This microbial dysbiosis can precede clinical signs of periodontitis, suggesting the importance of preventive strategies in periodontics. Probiotics have demonstrated potential in controlling the oral microbiota. To act effectively in this environment, they must adhere to dental surfaces, produce antimicrobial substances, reduce inflammation, and modulate the immune system. Studies report that strains such as Lacticaseibacillus rhamnosus GG are effective as adjuvants in periodontal treatment, with a positive impact on periodontal clinical parameters. Therefore, this study proposed the development of a hydrogel with L. rhamnosus GG, aiming at its physicochemical characterization and stability. The gel was formulated with low, medium, and high molecular weight hydroxyethylcellulose and the probiotic, following rigorous laboratory protocols. Data were statistically analyzed via ANOVA and non-parametric tests according to the data distribution. A statistically significant difference was observed in the stability of L. rhamnosus GG in relation to storage times T0 and T18 (p=0.002). No statistically significant differences were observed between the groups (p=1.000) in relation to appearance, color, and odor at the two times analyzed (day 0/day 90). The comparison of pH values between the different molecular weight groups showed similar averages between the evaluated formulations; no statistically significant differences were observed (p=0.209). Statistical analysis did not show a significant difference between the molecular weight groups and with or without the incorporation of the probiotic in relation to conductivity (p=0.081). Comparison of the average hydrodynamic diameter between formulations with and without the incorporation of the probiotic revealed statistically significant differences (p=0.028) and between the different molecular weights (p=0.042). A significant interaction effect was observed between the presence of the probiotic and the molecular weight of the polymer on the average hydrodynamic size (Z-average) of the formulations (p = 0.050). Comparison between groups did not reveal statistically significant differences in the polydispersity index values in the presence of the probiotic (p=0.054) and in the molecular weight of the polymer (p=0.677). Comparison of the zeta potential between hydrogels with and without the incorporation of Lacticaseibacillus showed a statistically significant difference between the groups (p=0.05). 3% hydroxyethylcellulose constitutes a suitable matrix for the incorporation of Lacticaseibacillus rhamnosus GG, maintaining physicochemical characteristics throughout the storage period of this study.
