Squamous cell carcinoma represents the most common malignancy in the mouth, accounting for more than 90% of cancers in this region. Among the most affected anatomical sites, the lower lip and tongue stand out, which commonly show different biological behavior, tending to be less aggressive with a higher survival rate and lower risk of metastasis when affecting the lower lip, when compared to oral tongue carcinomas that exhibit a more aggressive evolution and unfavorable prognosis. Therefore, it is important to search for markers that can indicate these differences and, in this sense, some studies have analyzed the role of the cells that make up the tumor microenvironment in the development and progression of oral carcinomas. Among these cells, tumor-associated macrophages stand out, which can present different phenotypes, with the M1 phenotype, represented by immunoeffector cells capable of destroying neoplastic cells, and the M2 phenotype, which has the ability to suppress the function of M1 macrophages, influencing the angiogenesis and tissue repair. The objective of the present study will be to evaluate the immunohistochemical expression of macrophage subpopulations in cases of squamous cell carcinomas located in the oral tongue (SCCOT) and lower lip (SCCLL), investigating possible associations with histopathological characteristics. To this end, 25 cases of SCCOT and 25 cases of SCCLL diagnosed at the Pathological Anatomy and Cytopathology service of the Department of Dentistry at UFRN will be selected for morphological and immunohistochemical study. The cases will be classified histologically using the grading system proposed by the WHO (2005) and subjected to semiquantitative analysis of the immunohistochemical expression of anti-CD68 and anti-CD163 antibodies, which mark, respectively, the M1 and M2 macrophage subpopulations. Associations between histopathological grading and macrophage subpopulations in different locations, as well as between locations, will be tested. The results obtained will be subjected to the Pearson chi-square parametric statistical test and the Mann-Whitney non-parametric test. It is expected that the results of this study indicate an association between macrophage subpopulations and the different histopathological grades of SCCOT and SCCLL. With this study, we intend to contribute to advances in understanding the role of these cells in tumor biological behavior.
