SUMMARY:

Thanks to its versatility and ability to produce complex structures with high precision, 3D printing has been increasingly used to manufacture scaffolds using biomaterials for studies and research. Polylactic acid (PLA) has been highlighted as a promising biomaterial due to its biocompatibility and biodegradability. However, PLA has some limitations, such as low bioactivity. To overcome these limitations, surface modification is a viable strategy, and using cold atmospheric plasma has demonstrated favorable results in other biomaterials. This project aims to evaluate whether surface treatment using the cold atmospheric plasma technique can modify the cellular response of fibroblasts and osteoblasts cultured on a PLA scaffold produced by 3D printing. The treated scaffolds will be compared to the untreated ones (control) concerning the following surface parameters: wettability (by the sessile drop test), morphology (by scanning electron microscopy - SEM), and nanotopography (by atomic force microscopy). After disinfection, cells from the commercial lines NIH-3T3 (fibroblasts) and MC3TE-E1 (pre-osteoblasts) will be cultured on the surfaces of the two groups. At intervals of 24, 48, and 72 hours after cultivation, cell viability and proliferation will be assessed by the Alamar blue assay, cell cycle events by flow cytometry, and events related to cell death by Annexin V/PI expression. The morphology of the cells on the treated and untreated surfaces will be analyzed by SEM. There is expected to be a significant increase in cell viability and proliferation in scaffolds treated by the cold atmospheric plasma technique, which can enhance the use of this tool in protocols involving polymeric biomaterials and tissue engineering.
