SUMMARY:

Amputation consists of the removal of a body segment or part of it, which can occur traumatically
or as the last available therapeutic resource in order to reduce the risks to the patient's life. Pain is
the most prevalent clinical manifestation in the postoperative period, around 70% of amputees
experience some type of pain and even at low intensity, it causes suffering, changing life in some
way. There is no consensus in the scientific literature on the most appropriate way to manage this
event, and there are numerous therapies. Within the scope of drug therapies, there are several
promising modalities and the analysis of evidence is important to support safe therapeutic choices
for the patient. Therefore, the study's general objective is to evaluate the effectiveness and safety of
using drug therapies for pain management in patients undergoing amputation surgery. This study is
an overview of a systematic review and network meta-analysis, to be conducted in accordance with
the Preferred Reporting Items for Systematic Reviews and Meta-Analyses for Network Meta-
Analyses (PRISMA-NMA). This overview will be registered in the International prospective
register of systematic reviews (PROSPERO) database. Using the “PICOS” strategy (Population,
Intervention, Comparison, Outcome and Type of Study), the research question was constructed:
“What is the effectiveness and safety of drug therapies when compared to placebos and/or other
treatments for pain management?” in patients undergoing amputation surgery?” The search will be
carried out by two researchers simultaneously in the databases: Catalog of Theses and Dissertations
(CAPES), Cochrane Library, Google Scholar, Latin American and Caribbean Literature on Health
Sciences (LILACS), National Library of Medicine and National Institutes of Health ( PUBMED),
Scientific Electronic Library Online (SCIELO), Science Direct, Elsevier's Scopus (SCOPUS), Web
of Science and Wiley Online Library. The Health Sciences/Medical Subject Headings Descriptors
will be used: (amputation OR surgical amputation OR traumatic amputation) AND (pain OR pain
management) AND (drug therapy) AND (systematic review). All Cochrane Systematic Reviews
(CSRs) and non-Cochrane Systematic Reviews (non-CSRs) with or without meta-analysis will be
included in the study, which described the population aged 18 years or over, with the use of
pharmacological therapies for the pain management in patients undergoing amputation surgery.
There will be no time restrictions for the selection of studies or the language of publications,
duplicate texts in the databases will be excluded. The results will be extracted using the Preferred
Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) flow diagram and a
standardized data extraction form containing information about the authors, year of publication,
study location, type of study, main objectives, population, age, sex, type of amputation, drug
therapy, placebo and other therapies used, route of administration, pain characterization, pain
assessment scales, and side effects. The network meta-analysis will be carried out using the R
software, considering 95% confidence intervals (CI). Methodological and reporting quality will be
assessed using Assessment of Multiple Systematic Reviews (AMSTAR 2).
