SUMMARY:
Trypanosomiasis are protozoa that occur in the Americas, highlighting the infection by
Trypanosoma cruzi and Leishmania spp. In this study, the occurrence of natural infection by
trypanosomatids in peridomiciled animals was evaluated using molecular markers and
phylogenetic analysis. The samples were blood clots from 163 animals: 60 sheep (Ovis aires),
22 goats (Capra hircus), 14 horses (Equus caballus), three cattle (Bos tauros) and 64 dogs
(Canis domiciliares) and 64 aspirated from popliteal lymph node of dogs in nine municipalities
in the state of Rio Grande do Norte (RN). Popliteal aspirate samples were added in culture and
the isolates were also analysed by molecular markers and phylogenetic analysis. Natural
infection by Trypanosoma spp. was identified by amplification of the 18S SSU rRNA gene in
48.9% of goat, sheep and equine samples. In the phylogenetic analysis, the SH022 sample
showed 99.8% similarity with the Y strain of T. cruzi grouping in the DTU II clade. Clot
samples from sheep, goats and horses amplified T. cruzi kDNA by 28.3% (17/60), 22.7% (5/22)
and 15.4% (2/14), respectively. These samples were distributed in the three studied
mesoregions. Data with dog samples revealed that popliteal cultures in biphasic medium were
12.5% (8/64) of the positive samples with flagellated forms similar to promastigotes, preceded
by the cities of Marcelino Vieira (3/6), Alexandria (3 /3) and Christmas (2/7). Blood clot
samples from dogs were also submitted to T. cruzi kDNA amplification and showed 10.9%
(7/64) positivity. In addition to the clots, the DNA from the wet mass of parasites from the eight
positive samples in the cultures was submitted to the same amplification showing bands of
650bp. In the sequencing and phylogenetic analysis, all samples of the culture showed similarity
above 99% with the Leishmania chagasi sequences. The wide distribution of T.cruzi and
Leishmania chagasi infection in domesticated animals contributes to the expansion of the
epidemiological scenario and the need to better understand the role of these animals in the
transmission cycle as reservoirs and/or sentinels of infections in humans.
