SUMMARY:
In the last decades, healthcare faces a scenario of shortage in the production of new
antibiotics and the increasingly frequent presence of infections caused by multidrug
resistant bacteria, making treatment a great challenge. Given this scenario, several
studies have been dedicated to the search for therapeutic alternatives, among them
are antimicrobial peptides (PAMs). TsAP-2 is a multifunctional PAM with
antimicrobial, antifungal, antiparasitic, antiviral, antiproliferative, immunomodulatory
and healing activities, present in the venom of the scorpion Tityus stigmurus. It has a
primary sequence - FLGMIPGLIGGLISAFK-NH2, 2+ charge and amphipathic
character, predominantly structured in α-helix. TsAP-2 stands out for its antimicrobial
activities with greater selectivity in Gram-positive strains. The association of
antibiotics is a common practice, which promotes the potentiation of antimicrobial
activity, elimination of multidrug-resistant strains, dose reduction, toxicity and risks of
resistance development. Therefore, the objective of this work was to evaluate the
association of TsAP-2 with conventional antibiotics in multidrug-resistant strains of
Staphylococcus aureus, using the checkerboard method. As well as, to evaluate the
antibiofilm and cytotoxic effect of the peptide in vitro and in vivo, in larvae of Galleria
mellonela. In conclusion, TsAP-2 showed activity against all multidrug resistant S.
aureus strains tested, significant activity on early and low biofilm on late multidrug S.
aureus biofilm, and had additive and synergistic effects in 50% of the associations,
where he obtained the best results combined with β-lactams. TsAP-2 had a high
hemolytic effect as a disadvantage, despite the low cytotoxicity in mammalian cells
(VERO). While, in in vivo tests, the peptide showed preservation of its antimicrobial
activities, revealing a 50% reduction of circulating bacteria in the hemolymph and
promoted an increase in larval survival, with low toxicity.
