SUMMARY:

The species Erythrina velutina Willd., Popularly known as “mulungu”, belonging to the
Fabaceae family and Papilionoideae subfamily, is distributed throughout the Northeast of
Brazil. Its leaves, seeds and mainly the bark of trunks are used in folk medicine in the form of
teas for the treatment of sleep disorders and disorders of the central nervous system (CNS).
Previous works with the species relate its pharmacological, anxiolytic and sedative properties,
mainly to the presence of flavonoids and benzylisoquinoline alkaloids. In a previous stage of
the work, our group carried out integrative studies of data from the transcriptome, proteome
and metabolome of this species, collected in four different locations in the Seridó region of Rio
Grande do Norte. The study of the targeted metabolic profile allowed the characterization of
the alkaloids present in this species through the use of molecular networking tools. Thus, the
present study aimed to continue this characterization, however, focusing on the phenolic
compounds present in the species. In order to facilitate the characterization of chemical
structures of special metabolites present in plant material based on mass data and mass
fragmentation patterns by spectral similarity, the de-replication of crude extract from samples
from different populations by liquid chromatography coupled with sequential mass
spectrometry (LC-MS/MS) with the help of the GNPS platform (Global Natural Product Social
Molecular Networking), led to the formation of a molecular network that presented three
clusters attributed to phenolic compounds, with the total number of 72 compounds, 32
flavonoids and 2 coumarins annotated by the GNPS platform library. The analysis of the nonannotated
metabolites indicated similarities in the fragmentation pattern which can be related
to the aglycone present in the sample with some variations present in the substituents.
Complementary and confirmatory analyzes using gas chromatography were initiated by
performing the extraction with seed extracting solvent and obtained a yield of 0.9615g for
ethanol and 0.8063g for methanol to standardize the analysis procedure that will be used in
samples from different populations. The next steps include the completion of the notes from the
comparison with the mass spectra present in the literature and the development of a protocol
for gas chromatography analysis coupled with mass spectrometry (GC-MS) of samples from
different populations of E. velutina aiming at characterization of more volatile phenolic
compounds.
