Accidents caused by scorpions, especially Tityus serrulatus, represent a serious public health problem due to the frequency with which they occur and the mortality they cause, especially in children, in several regions of Brazil. T. serrulatus or yellow scorpion as it is commonly known, is considered the most poisonous in South America due to the high toxicity of its venom. The main clinical manifestations can be: pain, vomiting, nausea, diarrhea, intense sweating, agitation, acute pulmonary edema, cardiac alterations and tremors due to natriuretic peptides, high molecular weight proteins, hyaluronidases, metalloproteiases, serine proteiases, peptides and proteins of low molar mass with neurotoxic activities, which are part of the constitution of the venom. The only form of therapy scientifically validated is antivenom serum therapy, which, however, has limitations such as serious hypersensitivity reactions to patients, difficult access and high cost. Therefore, the search for new alternatives to serum therapy becomes important, and in this context, many medicinal plants have been highlighted by popular use. Among these plants, we can mention the species Jatropha mollissima (Euphorbiaceae) that presents wide popular use in traditional medicine as antiofidic, anti-inflammatory, antimicrobial and antipyretc. The aim of this work was to evaluate the activity of the aqueous extracts of the leaves of J. mollissima in inhibiting the inflammatory and toxic effects induced by carrageenan and the venom of the T. serrulatus, respectively. The leaf extract was prepared by decoction. The antiinflammatory activity of the extract was evaluated in models of paw edema and peritonitis induced by carrageenan. The antiscorpionic activity of the extract was evaluated through inhibition of lung edema, myeloperoxidase enzymes, proinflammatory cytokines and serum levels of enzymes and proteins induced by T. serrulatus venom (TsV), with Swiss mice of both sexes being used.
In all models, the extract was evaluated orally at doses of 100 and 200 mg/Kg, and the protocol of pre and post-treatment was used only in experiments with the venom of TsV. All tested doses of the J. mollissima extract reduced paw edema with intensity similar to dexamethasone induced by carrageenan. The aqueous extract of J. mollissima, at all doses evaluated, inhibited cell migration induced by carrageenan. Pulmonary edema induced by TsV was significantly inhibited by the extract. The activity of MPO and the expression of IL-6 and IL-1β cytokines were reduced in the presence of the extracts significantly when compared to TsV. LDH, CK, Urea, Creatinine, AST, and Amylase levels were decreased by the extract of J. mollissima. These results indicate that the aqueous extract of J. mollissima presents potential anti-scorpion properties, especially in relation to inflammatory activity, which could justify the use of this plant in traditional medicine and complementary therapy as anti-scorpin activity.
