RESUMO:

The chitosan (QUI) and guar gum (GG) particulate systems for colonic transfer and controlled release of triamcinolone (TRI) were prepared using the simple coacervation technique with the addition of sodium sulphate followed by the spray drying drying method. Pre-tests were standardized, the inlet temperature of 140 ° C and outlet 90 ° C, blower 6.5 and atomization of air 0.1 ppm. Different drug-polymer ratios (1:5, 1:10 and 1:20) and polymer-polymer (50:50, 25:75 and 75:25 w / w) were studied for yield (%), encapsulation efficiency EE (%) and loading (mg TRI / g SP), infrared, DSC, TG, kinetics, swelling and in vivo study to optimize the preparation conditions of the obtained particulate systems. The particulate system (75:25 w/w) with a higher percentage of the chitosan to gum showed better yield (%), however, the particulate system (25:75 w / w) has a higher capacity of encapsulation efficiency (%), loading (mg TRI / g SP) and lower mass loss in the thermal analysis (TG), but due to its low yield, it was chosen to use in the in vivo study the polymer ratio 50:50 w / w with triamcinolone 1:10. Because this sample shows a good yield and a reasonable encapsulation efficiency, and still taking into account the necessity of the swelling and mucoadhesion properties. The characterization of the infrared systems evidenced the interactions between the components of the formulation. The thermal analysis showed that the systems with greater amount of chitosan presented a greater thermal stability. The kinetic study of the three polymeric ratios (QUIT: GG 25:75, QUIT: GG 50:50 and QUIT 75:25) with triamcinolone 1:10 occurred in two phases, the first with a rapid initial release (burst effect) in 1 hour, releasing from 19% to 27% of the encapsulated drug for the different formulations. The second phase of release is characterized by being more controlled, being attributed to the diffusion of the drug through the polymer matrix, after 24 hours almost 100% of the samples were released. The swelling study showed a statistical difference of 24h to 48h, with p <0.05, with tabulated F (5.14) << F calculated (79,14), the polymer ratio 75:25 w / w, more amount of chitosan is swelling more than the other reasons. All the particulate systems (25:75, 50:50 and 75:25) w / w are swelling more than dry placebos by spray drying and by physical mixing, in the first 9 hours it is possible to observe that SP gain more mass (P <0.05), with the tabulated F (3.47) << F calculated (15.86), remaining higher until the end of the 48h analysis. However, the physical mixing placebo had a greater degree of swelling than dry placebo by spray drying. Therefore, a new technological system was obtained by drying in spray drying of particulate systems based on chitosan and guar gum capable of modifying the release of triamcinolone with potential for application as anti-inflammatory in ulcerative colitis.
