SUMMARY:

Oral squamous cell carcinoma (OSCC) is the sixth most common type of cancer worldwide and great efforts are made to identify prognostic factors (clinical, morphological and molecular) that can best predict patient survival and that potentially stratifies patients into individualized therapeutic options. In this perspective, there is the heat shock factor 1 (HSF1), considered the main activated transcription factor in response to proteotoxic stress, a common event in cancer cells. Studies in several human tumors provide increasing evidence that HSF1-dependent regulation is involved in various processes associated with tumor initiation and progression, as well as therapeutic resistance. Therefore, clarifying the pathways of HSF1 participation in OSCC may help identify the functions of this factor in the biological behavior of this tumor. Accordingly, this research aims to evaluate the participation of HSF1 in OSCC tumorigenesis through experiments with the cell line SCC15, silenced and non-silenced with lentiviral particles for the HSF1 gene, with silencing confirmed by qRT-PCR and Western Blot. We are going to analyze cell viability and proliferation (CellTiter and BRDU, respectively), necrosis and apoptosis levels (Annexin-Annexin-V-FITC and flow cytometric analysis), invasion and migration capacity (transwell/Matrigel system), influence on cell cycle (propidium iodide and flow cytometric analysis) and epithelial-mesenchymal transition (expression of E-cadherin and vimentin quantified by qRT-PCR). All experiments will be repeated in triplicate and for all statistical tests used in this study the significance level will be 5%.
