RESUMO:

Gingival inflammation due to colonization by periodontopathogenic microorganisms associated with host response generates immunological and vascular events due to the release and activation of inflammatory mediators. This inflammatory response varies according to the age of the patient, making healing and repair of the region difficult. In this perspective, this study aims to evaluate cells of the innate immune system and the presence of blood vessels in 180 specimens of gingival tissue classified as gingival health, gingivitis induced by biofilm, periodontitis stages III and IV, in grades A, B and C, being considered the age of the patient, divided into young and old adults. Morphological analysis under optical microscopy using hematoxylin and eosin staining to evaluate the characteristics of epithelium and connective tissue present. In the immunohistochemical analysis will be used the CD1a immunomarkers, for immature dendritic cells; CD83, for mature dendritic cells; tryptase for mast cells; and CD34 for endothelial cells. The CD1a, CD83 and Triptase immunoresponsors will be evaluated by labeling density in five areas of greatest immunostaining in the specimen, where the number of positive cells in each selected area will be quantified, and the mean number of marked cells in the 5 fields will be quantified. CD34 will be evaluated through the angiogenic index, where the microvascular count, microvascular perimeter and microvascular area of five areas with greater immunolabeling will be performed. The program to be used for these analyzes will be Image J. The clinical, morphological and immunohistochemical data will be tabulated in Microsoft Excel® program tables and later exported to the statistical program SPSS® version 20.0, in which they will be subjected to a descriptive analysis and inferential, probably through non-parametric tests. For all statistical tests to be used in this study, the level of significance will be set at 5% (p <0.05).
