RESUMO:

Oral mucositis (MO) is a sequela of frequent and severe oncological treatment, which may compromise the progress of the patient's cancer treatment, since it contributes to the debilitation of his general condition. Clinically, OM varies in degree of aggressiveness and can show from erythematous areas to ulcerations, which can cause pain and consequent food restriction, as well as being a site of secondary infection. Effective interventions for the prevention and treatment of MO should be priorities in support care for cancer patients. Propolis and royal jelly have been widely used in the treatment of wounds due to their cicatrizing, antimicrobial, anti-inflammatory, analgesic and antioxidant actions. Considering that studies with phytotherapics in the treatment of MO are scarce and that the development of new therapies for this condition is of great importance, since the oncological services are not always available, as well as the mechanisms of these drugs acting in the cicatrization of these lesions , the objective of the present study was to evaluate the clinical, histopathological and PI3K mTOR pathway participation in the use of propolis and royal jelly in 5-fluorouracil (5-FU) induced MO in rats, comparing the action of these compounds with the well-established action of Photobiomodulation (FBM) with laser. Wistar rats were randomly divided into 4 groups (each group with n = 18): 1 - negative control group (induction of untreated mucositis), 2 - positive control group (induction of mucositis and FBM use), 3 - propolis gel test group (induction of mucositis and use of propolis gel) and 4 - test group royal jelly (induction of mucositis and use of royal jelly). The animals received intraperitoneal injection of 5-FU on days 0 and 2. On days 3 and 4 the jugal mucosa was scarified, beginning treatment of OM on day 5. The animals received two daily applications of the product (according to their experimental group ) with a flexible rod in the area where mucositis was induced. The FBM group received only one daily application (Single point / 14.4 seconds / 6J / cm²). Analysis times occurred on days 8, 10 and 14 days after chemotherapy infusion. In these days, six animals from each group were euthanized, weighed, and the jugal mucosa was photographed for clinical grading of MO and removed for morphological and immunohistochemical analyzes. The fragments were fixed in 10% buffered formalin solution and embedded in paraffin for further histopathological (Hematoxylin / Eosin) and immunohistochemical analysis for pAKT and p-S6 antibodies. The initial descriptive analysis of the results was performed according to the experimental groups considering the treatments performed. Subsequently the statistical analysis will be performed considering the treatments and the levels of the chemical mediator at each time as main effects and the interaction time x treatment through the PROC MIXED of the SAS program. The multiple comparisons will be performed using the Kruskal-Wallis test (KW) adjusted for the correlations between the times.
