RESUMO:

Squamous cell carcinoma (SCC) accounts for 90% to 95% of oral cavity malignancies, mainly on the lateral border of tongue, and it is clearly represented as a public health problem. The causes are extrinsic and intrinsic multifocal factors which can cause genetic alterations, producing defective proteins, so compromise important processes such as DNA repair and cell cycle control. The base excision DNA repair pathway (BER) is responsible for DNA repair after oxidative damage and includes the APE1 and XRCC1 genes. The APE1 protein acts by increasing the binding activity to DNA transcription factors (fos, jun, nuclear factor-κB and p53). XRCC-1 acts in support of other repair factors and interacts with BER pathway proteins, including APE1. P53 is a tumor suppressor gene. Mutations in this and defective production of the p53 protein can cause uncontrolled cell proliferation and contribute to onset of cancer. Ki-67 is a protein that can be used as a marker for different cell growth conditions and cell proliferation is associated with progression and recurrence in human cancers. Few studies are available on the role of DNA repair gene expression and its association with proliferation and carcinogenesis. The aim of this research is to analyze the immunohistochemical expression of DNA repair proteins (APE1, XRCC1) and proteins involved in the process of cell proliferation (p53 and ki67), associating them with clinical and histopathological parameters in CCEO of tongue, aiming to contribute for the better understanding regarding the participation of these proteins in the development and evolution of this neoplasia.
