RESUMO:

Low-level laser irradiation (LLLI) has been used to promote wound healing and tissue regeneration, but little is known about their effectiveness in the proliferation and differentiation of stem cells from dental origin. The study aims to evaluate, through in vitro and in vivo experiments, the influence of LLLI on the biological activity of stem cells from human exfoliated deciduous teeth (SHED). Dental pulps will be obtained from six primary teeth in the final stage of exfoliation and the SHED will be expanded. In the in vitro experiment, cells will be irradiated with two doses (0.5 and 1.0 J/cm2), with a non irratiated group as control. At intervals of 0, 24, 48, and 72 h after the first irradiation, cell proliferation will be assessed by trypan blue exclusion method and MTT assay, and viability and cell cycle evaluated by flow cytometry. The effect of LLLI on in vitro cell differentiation will be evaluated by identifying mineralized nodules in culture plates, as well as by alkaline phosphatase and osteocalcin expression by flow cytometry. In the in vivo trial SHEDs associated with ceramic particles of hydroxyapatite and tricalcium phosphate (HA/TCP) will be grafted in the subcutaneous tissue of C57Bl/6 mice. The animals will be divided into three groups, one control and two submitted to laser therapy with doses of 0.5 and 1.0 J/cm2 every other day for 15 days. After 4 weeks, areas of transplants will be subjected to evaluation of tissue parameters (inflammatory infiltrate, vascular neoformation, degree of collagenization and formation of mineralized tissue) and analysis of alkaline phosphatase and osteocalcin expression by Western blotting. Quantitative data will be submitted to non-parametric statistical analysis and the descriptive data will be used for comparison between groups.
