SUMMARY:
Jambolan (Syzygium cumini) fruits are rich in anthocyanins and other phenolic
compounds, molecules recognized for their high antioxidant potential, but whose
chemical instability demands well-defined extraction processes. Thus, the present
study investigated different solid-liquid extraction techniques (Soxhlet, ultrasound
assisted bath, and pressurized liquid extraction) combined with different solvents
(ethanol and methanol) and extraction times, seeking to maximize the recovery of
these bioactive compounds. From the initial screening, the extract obtained by
pressurized liquid extraction (PLE) stood out as the most promising, reaching TMA
levels of 17.92 mg·g-1 and TPC of 897.73 µg GAE·g-1, in addition to an ABTS+ radical
scavenging activity of 2.38 µmol TE·g-1 and TAC of 220.12 mg AA·g-1. Subsequently,
the phytochemical profile was characterized by HPLC-DAD, and in vitro cytotoxicity
was evaluated. A total of 38 compounds were identified, highlighting pelargonidin-3,5-
diglucoside (114 µg·g-1) and cyanidin-3-glucoside (112 µg·g-1), followed by peonidin-
3-glucoside (11 µg·g-1) and malvidin-3-glucoside (10 µg·g-1), as well as gallic acid (711
µg·g-1), procyanidin A2 (201 µg·g-1), 4-hydroxybenzoic acid (127 µg·g-1), and
epigallocatechin gallate (101 µg·g-1). Regarding cytotoxicity in the HepG-2 cell line, cell
proliferation was stimulated (1000 μg·mL-1 at 24 h and at all concentrations at 72 h),
with cell viability values ranging from 121.6% to 133.1%. In CHO-K1 cells, no
cytotoxicity was observed at any concentration at 24 h, nor up to 500 μg·mL-1 at 72 h.
The use of PLE allowed for the extraction of a robust phytochemical matrix from S.
cumini, paving the way for its potential use in future technological and pharmaceutical
applications.
