In recent scientific research, degenerative diseases, which cause long, arduous and costly treatments (due to the use of high doses of therapeutic drugs) to patients with chronic diseases, are the focus of studies for researchers in various scientific areas. As a colossal alternative to increasing therapeutic efficacy drugs, providing a comfortable and convenient treatment for patients, especially those who make frequent use of therapeutic drugs, due to a disease of chronic origin, drug delivery systems (drugs delivery systems) have become an excellent option for this process of improving the clinical response of patients with certain diseases. Epilepsy is a neurological disorder with a higher occurrence in the world population. Adverse reactions related to treatment neurological diseases, in this case epilepsy, occur due to the use of high dosages of antiepileptic drugs and a use generally throughout life. Topiramate is a drug used to treat epilepsy, including Lennox-Gastaut syndrome, and other neurological disorders. Patients who use Topiramate as an antiepileptic drug need up to 1600 mg of the drug daily in several doses throughout the day. Thus, development of materials for storage and modified release of drugs has been highlighted in the scientific area. This term modified, sustained, slow or sustained release refers to the release of the drug into the body gradually, keeping the drug within therapeutic levels. The present study was based on the principle of silica nanoparticle synthesis (NPS), using a natural source of silica as precursor (a expanded perlite, natural, low-cost, abundant aluminosilicate silica-rich). Nanoparticles were used as inorganic matrices for the storage and transport of antiepileptic topiramate in controlled release studies in vitro. The parameters of the modified Stöber method were varied to obtain particles of different sizes and size distributions (monomodal and multimodal), in particular the monomodal NPS of different sizes (548.3 nm, 525.7 nm and 484.4 nm ), which were characterized by Dynamic Light Scattering (DLS), XRD, FTIR and Nitrogen adsorption / desorption (BET method). The drug incorporation study was performed in NPS, varying the parameters of the concentration of the Topiramate solution and the nanocarrier mass (NPS), in which an incorporation of 80.0% of the dose of the drug was achieved in the NPS. All the trials in which it was necessary to detect or quantify Topiramate, the CG-MS technique was used, whose analysis methodology was optimized and defined for the detection and quantification of the drug through several previous tests with Topiramate 98.0% of purity, used as the standard in this study. The analytical validation parameters for the methodology of analysis of Topiramate by GC-MS were obtained (limits of quantification and detection) according to the current RDC No. 166 (2017) of ANVISA. Thus, NPS has been shown to be a promising material in modulating the concentration of Topiramate in this silica carrier, and will be used in in vitro drug release studies, which shows a relevant study to reduce the high dosages of Topiramato and numerous reactions adverse effects that accompany patients who use this antiepileptic, and may also reduce the financial cost and bring more quality of life to those who are treated with this antiepileptic that is usually for a lifetime.
