RESUMO:

The aim of this present work consist in the preparation of microemulsion systems based on clove oil (Syzigium aromaticum L.) to be applied on health care as part of biotechnological development of new microstructured products. The clove oil standard formulation (so called SEWOCR) was prepared by using a fixed amount of nonionic surfactant, the oil phase ranging from 2% to 3% (OF) and the aqueous phase varying from 83%  to 84%. The minimum amount of surfactant required to prepare the SEWOCR formulations was determined through limiting dilution concentration (LDC) study in bidistilled water. The oil phase varied according to the origin of the clove oil, such as: clove oil extraction at room temperature using maceration method, Soxhlet extraction or a clove oil commercial sample. From the Winsor IV microemulsion region in the ternary diagram of the clove oil standard formulation (so called SEWOCR) it was prepared some derivative systems (SEWOCR1a, SEWOCR2, SEWOCR2a and SEWOCR3a) which physicochemical properties were characterized by using surface tension, viscosity, droplet diameter, density and zeta potential. The surface tension data showed no significant variations among the SEWOCR derivative systems. However,  the formulation containing 3% of of the oil phase (OF), significant elevations in the values of surface tension was observed, in which the LDC was 0.0015 g/mL) and the maximum number of dilutions (MD) was 27.26. Meanwhile, formulations with lower amount of the OF, sowed LDC 0.0010 g/mL and MD of 49.07. Proportional to the OF increased concentration it was observed 103.8 cP as maximum value for the formulation containing 3% of OF, and for the other SEWOCR derivative systems lowest data ranging from 1.9 cP to 15 cP were evidenced. Comparatively, the droplet analyses showed similar behavior with maximum value 25.61 nm, for the  formulation containing 3% of OF and data ranging from 9,55 nm to 16,54 nm for the other systems with lowest OF content. In the other hand, for all tested systems  similar values were observed in the analyses of zeta potential which varying only in the third decimal order. According to the physicochemical characterizations, it was observed that, regardless of the oil origin the formulations containing lower OF concentrations present the more satisfactory results, such as: low surface tension and viscosity, reduced droplet diameter value as well as LDC content. The phytochemical characterization of the used clove oils was done via CG-MS analyses and results showed variation in the content of the following components: eugenol (63.75% - 61.36%), β-caryophyllene (16.98% - 14.21%) and acetyl eugenol (10.61% - 9.54%), been in accordance with the oil origin. Quantification of the biomarker eugenol in the SEWOCR microemulsion systems was performed via RAMAN spectroscopy. The pharmacological efficacy of the SEWOCR1a, SEWOCR2, SEWOCR2a and SEWOCR3a systems was evaluated by using in vitro experiments such as antioxidant tests and by the viability of dental pulp stem cells (Dental PulpStemCells), and also by applying an in vivo experiment for analgesic effect evaluation.  s well as in, and effective action was observed. The antioxidant efficacy was proven in the tests of total antioxidant activity, reducing power and sequestration of hydroxyl radicals. The analgesic effect via essays of abdominal contortions induced by acetic acid, showed the antinociceptive activity.
