Cancer and bacterial resistance are among the main currently challenges in the biomedical field, this scenario requires the development of alternatives for treatment. During the doctoral period are proposed the synthesis and characterization of hybrid systems containing nanoparticles of noble metals (Ag and Au) and drugs for antibacterial and antineoplastic application. The suggestion of a biochemical route of action of these systems in bacterial cells is also part of the proposals. For qualifying examination will be presented the synthesis and characterization of gold nanoparticles (NPsAu) and biological assays before the combination with drugs. NPsAu are considered as potential drug delivery platforms. However, it is essential to understand the interaction of these particles with living organisms prior to their use as nanocarriers. For this purpose, anti-inflammatory, analgesic and antitumor activity of NPsAu and biodistribution thereof and their effect on various tissues were studied. The NPsAu were synthesized through an low environmental impact route and characterized with transmission electron microscopy, TEM and spectroscopy in the UV-visible region, UV-Vis. Later HT-29 cells were exposed to NPsAu and then, apoptosis and caspase-3 activity was evaluated. Concerning to the in vivo essays, the NPsAu were administered to male and female swiss mice and rats for later evaluating their anti-inflammatory and analgesic properties. The biodistribution of NPsAu and its impact on the tissues were studied by UV-Vis and histopathology analysis, respectively. Apoptosis of the cells was observed a dose-dependent manner, to NPsAu concentrations ranging from 40 mg ml-1 to 80 mg ml-1 (p <0.05). The best anti-inflammatory activity was observed at a dose of 1500 mg kg-1, which caused a reduction of 49.3% in the migration of leukocytes. NPsAu showed peripheral analgesia in a dose of 1500 mg kg-1 and were found in the liver, spleen, kidney and lung of the studied animals. Histopathological examination revealed extravasation of red blood cells in the lung. Thus, it is possible to highlight the NPsAu obtained in this study as a resource for technological innovation in the fields of anti-inflammatory, analgesic and antitumorial action. Gold nanoparticles have biological effects that deserve investigation for the determination of their total interaction with biological systems.
