SUMMARY:
Snakebites is considered a relevant public health problem, included by the World
Health Organization in the list of Neglected Tropical Diseases. The most clinically
relevant genus, responsible for the largest number of accidents, is Bothrops. Common
events such as local edema, myotoxicity, hemorrhage and dermonecrosis characterize
accidents and can lead to severe damage to patients. The low efficiency of antivenom
serum therapy in the face of these events encourages the search for complementary
therapies. Medicinal plants have been used for years in traditional medicine in snakebites.
The plant species Ipomoea pes-caprae (Ipc), popularly known as “salsa-da-praia”, is used
in traditional medicine to treat inflammation, pain, burns and snake bites. Thus, this study
focused on the development and evaluation of formulations containing Ipc extract
regarding their antiophidic potential. The hydroethanolic extract of Ipc was obtained by
maceration. The results suggested by Thin Layer Chromatography and confirmed by the
analysis by High Performance Liquid Chromatography identified the majority presence
of phenolic compounds in the extract. Obtaining the fraction enriched with phenolic
compounds (FrEtOH) was successfully performed. The extract was incorporated into a
gel previously developed by the research group. After 90 days of the accelerated stability
study, the formulations with the extract were stable in terms of organoleptic and
physicochemical properties, as well as the phenolic content with variation according to
the legislation. The Ipc and FrEtOH extracts showed no in vitro cytotoxicity against
murine macrophages (RAW 267.3) and mouse embryonic fibroblasts (NIH/3T3) cells.
The in vitro enzymatic activities against Bothrops erythromelas (Ber) venom toxins
(phospholipase, proteolytic and hyaluronidase) were similarly inhibited by the Ipc extract
and FrEtOH. The in vivo activities against the local effects produced by the Ber venom
(edematogenic, hemorrhagic and dermonecrosis) were reduced by the treatment with the
Ipc gel 5%, as well as a synergistic effect when using the Ipc gel associated with the
antivenom. The inhibitions observed were promising, presenting a direct action of the
extract inhibiting venom toxins and an indirect action through the inhibition of
endogenous mediators involved in envenomation. In addition, it is possible to relate the
activity to the compounds present in the extract, with emphasis on the phenolic
compounds that act strongly as anti-inflammatory, antioxidant and metal chelator. Given
this scenario, the results point to the potential of the developed formulation, which may
justify the medicinal use of the extract in snake bites, and may also contribute to the
development of new herbal products on the market for the treatment of the local effects
of bothropic envenomation.
