SUMMARY:

Atmospheric pollution, originating from natural or anthropogenic sources, represents a serious global health problem. Biomass burning, one of the sources of such pollution, releases toxic compounds such as polycyclic aromatic hydrocarbons (PAHs). Among the PAHs, retene (RET) stands out as a compound that, although not included in the risk assessments of the United States Environmental Protection Agency (US-EPA), shows evidence of genotoxicity, cytotoxicity in human lung cell lines, and behavioral alterations in adult zebrafish. However, its impact on telomeres remains unknown, which is relevant because telomere shortening is a marker of cellular stress, aging, and increased risk for chronic diseases such as cancer. Thus, the aim of this project is to evaluate the effect of retene on telomere length in human bronchial epithelial cells (BEAS-2B) and in human lung adenocarcinoma cells (A549). The cells will be cultured and exposed to RET at three concentrations (10, 30, and 60 ng/mL) for 24 h. Telomere length will be analyzed using the quantitative polymerase chain reaction (qPCR) assay. In addition, cytokinesis-block micronucleus (CBMN) tests and the Comet assay will be employed to assess genotoxic damage. It is expected that this study will elucidate the mechanism of action of RET and its relationship with genomic instability, contributing to the assessment of its toxicological risk.
