Effect of Spondias mombin L. gel on inflammation, oxidative stress, and alveolar bone loss in rats subjected to experimental periodontitis.
Spondias; Phytotherapy; Periodontal disease.
The search for new products offering good therapeutic activity, greater tissue biocompatibility, and reduced adverse effects and toxicity has increasingly stimulated research into herbal medicines. The species *Spondias mombin* L. has gained prominence due to the anti-inflammatory, antimicrobial, and antioxidant effects of its leaves. The objective of this study is to evaluate the effects of a *Spondias mombin* L. gel on inflammation, oxidative stress, and alveolar bone loss in rats subjected to experimental periodontitis. This *in vivo* preclinical trial will assess the therapeutic action of a gel formulated from the hydroethanolic extract of *Spondias mombin* L. leaves (at concentrations of 1% and 3%) as an adjunct to non-surgical periodontal treatment (scaling and root planing) in 67 male Wistar rats, distributed into five experimental groups (healthy, untreated, scaling only, Periodontal Disease + 1% *Spondias mombin* gel, and Periodontal Disease + 3% *Spondias mombin* gel). Periodontitis will be induced by placing a 3.0 nylon ligature around the upper left second molar for 11 days; following ligature removal and scaling, the gels will be applied topically once daily for 14 consecutive days. The physicochemical stability and phytochemical profile (focusing on the markers chlorogenic acid and isoquercitrin) of the formulations will be monitored via macroscopic analysis, pH determination, total phenolic content analysis, thin-layer chromatography (TLC), and high-performance liquid chromatography with a diode-array detector (HPLC-UV/DAD). Following euthanasia, gingival tissues will be used to analyze malondialdehyde (MDA) levels and the expression of bone morphogenetic protein (BMP-2), runt-related transcription factor (RUNX), and osteocalcin (OC). The jaws will undergo histological, immunohistochemical, and micro-computed tomography analyses. The data obtained will be analyzed using descriptive and inferential statistics. Parametric tests (such as ANOVA followed by the Bonferroni post-test) and non-parametric tests (such as the Kruskal-Wallis test) will be employed, with a significance level of 5%.
