SUMMARY:

Giant cell lesions (GCLs) comprise a heterogeneous group of pathological entities characterized by the presence of osteoclast-like multinucleated cells, including Central Giant Cell Lesion (CGCL) and Giant Cell Tumor (GCT) of long bones. Although these lesions share histopathological similarities, they differ in terms of biological behavior and underlying molecular mechanisms. CGCL is a benign, non-odontogenic osteolytic lesion that predominantly affects young individuals and may exhibit variable clinical behavior, ranging from indolent forms to locally aggressive presentations with potential for bone destruction and recurrence. In contrast, GCT is considered a benign neoplasm with locally aggressive behavior and a high recurrence rate, occurring predominantly in young adults. Elucidation of the molecular mechanisms underlying the behavior of these lesions is essential for a better understanding of their pathogenesis. In this context, the Hippo signaling pathway, particularly the YAP/TEAD axis, has emerged as a key regulator of cell proliferation, differentiation, apoptosis, and tumorigenic processes. Activation of this axis has also been associated with the induction of transcriptional mechanisms involved in tissue reorganization and extracellular matrix remodeling, processes in which matrix metalloproteinases, such as MMP-7 and MMP-9, play a significant role, suggesting a potential association with the biological behavior of GCLs. Thus, this study aims to investigate the immunoexpression of YAP-1, TEAD1, MMP-7, and MMP-9 in central giant cell lesions (CGCLs) of the jaws, classified as aggressive and non-aggressive, as well as in giant cell tumors (GCTs) of long bones. This will be conducted through a semi-quantitative analysis of cellular localization and immunostaining distribution, followed by descriptive and inferential statistical analysis (p ≤ 0,05), aiming to correlate molecular findings with clinicopathological parameters to enhance prognostic stratification and therapeutic guidance.
