SUMMARY:

Arapaima gigas, known as pirarucu, is considered one of the largest freshwater fish in the world, with a growing interest in the aquaculture market due to its particular biological characteristics, including its rapid growth in its early years. In recent years, despite the massive availability of data from sequencing projects, few have addressed the taxon that includes this species. By cDNA sequencing of 12 different tissue samples from Arapaima gigas, we assembled a reference transcriptome that combines the new and genome-guided assembly strategies. The gene expression of gender between different male and female tissues of adult specimens was analyzed. We used pipelines such as Hisat2, Braker2, Trinity, Diamond and mygene for the assembly and annotation of genes. The present study was developed with the purpose of providing a final transcriptome, categorizing through a transcriptional exploratory analysis the specific gene expressions of processes related to sexing, gigantism, as well as highlighting specific tissue genes, unidentified genes and ancestral isoforms of genes. . In this study we highlight the discovery of genes not yet described in public repositories as well as a set of annotated gene products that may be candidates to affect sexual dimorphism in pirarucu, its size and accelerated development, and how ancestral candidate genes explain it’s unique morphological characteristics.
