RESUMO:

The present study aimed to evaluate the antioxidant and antiproliferative activity from Myrciaria tenella (DC.) O.Berg and Salvia hispanica L. extracts. In order to do this, it was obtained different extracts using a serial extraction method (apolar to polar solvents) using fresh leaves of M. tenella and S. hispanica. Then, it was obtained the following extract: hexane (EHC and EHSh); chloroform (ECC and HSCS); ethanoic (EEC and EESh); methanoic (EMC and EMSh) and final aqueous (EAFC and EAFSh). Furthermore, an aqueous extract was separately made (EAC and EASh). The extracts were characterized phytochemically by CCD and UPLC, quantification of sugars, proteins and total phenolic compounds. Moreover, the antioxidant activity was evaluated using different in vitro and in vivo assays on the Caenorhabditis elegans model. Furthermore, it was also evaluated the antiproliferative activity using the cell viability assay (MTT) method on murine fibroblasts (3T3) and human cervical cancer cells (HeLa). In order to evaluate the cell death on a flow cytometer, then it was analyzed the effect of EEC, EAFC, EESh and EASh on HeLa cells. The data obtained here shown first that M. tenella and S. hispanica had phenolic and terpene compounds. Ursolic acid was identified in EHC, ECC, EHSh and ECSh; saponins in EEC, EMC and EAFC; routine in EEC and EMC; as well as pyrogallol in EHSh and EAFSh. The antioxidant assays showed that extracts from M. tenella and S. hispanica were able to donate electrons to the oxidized substrate and chelate transition metals (iron and copper) in vitro. The extracts EEC and EASh, at concentrations 1 and 10 mg / mL, reduced the basal levels of ROS in C. elegans, by 44 and 70%. However, they did not alter were able to change the sod-3 expression when compared to the untreated control. The antiproliferativa assay using cell lineage showed that the different extracts of M. tenella and S. hispanica reduced the HeLa cells viability by up to 80%. A reduction from 16-56% was observed at the lowest concentration analyzed -100 μg/mL. However, it was not observed any change at cell death by flow cytometry. No change in viability of HeLa cells treated with EEC, EAFC and EASh at 100 μg/mL. It was observed a necrosis cell death only for EESh at this same concentration. In summary, the leaves of M. tenella and S. hispanica may be a good source of antioxidant activity as well as be as antiproliferative agent, which, might be a potential for the phytotherapics development.
