RESUMO:

Sugarcane is monocot that has a huge economic importance as it is used for ethanol and sugar production. This plant may grow in tropical and subtropical regions. However, the abiotic conditions as drought, salinity, high temperatures may reduced its productivity. Considering this, the aim of this work was to study the effect of changes of gravity forces such as hypergravity and microgravity on sugarcane transcriptome. In order to make this, sugarcane plants were submitted to a VSB-30 flight with 06 min. of microgravity. When these plants were recovered leaves and roots were separated, RNA was extracted and sequenced using Illumina plataform. The sequence received was analyzed using different bioinformatics tools. The transcriptome analysis from leaves and roots allowed us to identify 1387 constitutive genes and tissue specific genes for vertical root (454 genes), horizontal roots (478 genes), vertical leaves (147 genes) and leaves horizontal (49 genes). The constitutive genes were analyzed using the DESeq, which identified 154 genes that were specific. On the other hand, the tissue specific genes were evaluated by AgriGO tool and it was observed using some bioinformatics tolls that sugarcane plants in vertical orientation had probably differential expression for genes associated to nitrogen metabolism. It was also analyzed the possible interactomic network and it was observed that the root tissue had possible a complex response involving different proteomic interaction. Interactoma tool showed the presence of 12 clusters for vertical roots and 13 cluster for horizontal orientation in roots. These networks identified suggested a possible relation to nitrogen metabolism and amino acid biosynthesis. This work allowed us to identifiy what may be affected in the transcriptome during the VSB-30 flight and the data presented here suggested possible gene/protein network that may be changed due to hypergravity and microgravity.
