Malaria is an infectious parasitic disease caused by protozoa of the genus Plasmodium and, in its severe forms, may progress to malaria-associated acute respiratory distress syndrome (MA-ARDS), characterized by intense pulmonary inflammation. Although murine models are widely used to study cerebral malaria, limitations remain regarding the standardization of experimental models for pulmonary malaria. Therefore, this study aimed to standardize the murine strain and parasitic inoculum for the investigation of MA-ARDS. BALB/c and C57BL/6 mice were infected with Plasmodium berghei NK65 using different parasitic inocula and evaluated for parasitemia, pulmonary cell migration and differential counts, histopathological alterations, clinical progression, and survival. The results demonstrated that, in both strains, parasitemia showed a similar progression and was independent of the inoculum used. Both strains developed clinical and pathological features compatible with MA-ARDS; however, BALB/c mice exhibited a more intense pulmonary inflammatory response, with increased cellular recruitment and more extensive histopathological damage, whereas C57BL/6 mice showed a more moderate inflammatory profile and delayed clinical progression. Thus, BALB/c mice are more suitable for studies focused on exacerbated pulmonary inflammation and tissue damage, while C57BL/6 mice are more appropriate for investigations addressing inflammatory modulation and disease progression in MA-ARDS. Within the evaluated range, the parasitic inoculum was equally effective for model induction.
