Zinc and iron are essential for a large number of physiological processes in humans. The potential risk of interactions between zinc and iron that affect absorption and bioavailability has to be considered in any supplementation or fortification strategy. Thirty healthy children aged between 8 and 9 years were studied with the aim of verifying the effect of a 3-mo oral zinc supplementation on iron status. Fifteen individuals were given a placebo (control group), and fifteen were given 10 mg Zn/day (experimental group). Before zinc supplementation, all schoolchildren were submitted to venous zinc administration (0.065 mg Zn/kg of body weight). Blood samples were collected at 0, 60, 120, 180 and 210 min after a 12-h overnight fast. A significant increase in serum zinc during venous zinc administration was observed between the control and experimental groups after oral zinc supplementation or placebo (p<0.05). However, baseline serum iron concentrations significantly decreased in comparison to the control group (p<0.05) and remained at the same level throughout venous zinc administration. This result suggests that the acute infusion of zinc had no effect on serum iron. We ascribe the decrease in iron to the effect of chronic oral administration of this micronutrient. Concentrations of hemoglobin, mean corpuscular volume, ferritin, transferrin, transferrin saturation, ceruloplasmin and total proteins were in normal reference ranges. These results indicate that physiological doses of zinc administered for 3 mo can inhibit iron absorption in healthy children and that this antagonistic effect could compromise long-term iron status.
