Extraction of carotenoids from buriti peel and pulp (Mauritia flexuosa L.) using green solvents and evaluation of its bioactive properties

SUMMARY:

The buriti (Mauritia flexuosa L.) is a fruit that provides large amounts of bioactive components such as carotenoids and vitamin A. Thus, buriti represents a potential raw material for the development of cosmetic formulations due to the presence of antioxidant phytochemicals. In this sense, the objective of the present study was to extract the carotenoids from the buriti peel and pulp using green solvents for incorporation in cosmetic nanoemulsion. Initially, extractions of buriti peel and pulp were performed using deep eutectic solvents (DES), ethanol and supramolecular solvent (SUPRAS). In addition, the yield of carotenoid extraction with ethanol was evaluated by conventional technique and assisted by ultrasound. The concentration of total carotenoids in the extracts obtained was evaluated by spectrophotometry, and the concentration of total phenolic compounds and beta-carotene was determined by high-performance liquid chromatography (HPLC). The bioactive properties of the extracts were evaluated for in vitro antioxidant activity (DPPH and ABTS methods) and antibacterial activity by the broth microdilution method (MIC). The use of deep eutectic solvents showed satisfactory results when employed as adjuvant in the extraction process of total carotenoids, with a yield of 10.15 ± 0.25 mg/g for peel, and 10.30 ± 0.08 mg/g for pulp. The supramolecular solvent also showed excellent yield in the process of extracting beta-carotene from buriti pulp (26.70 ± 0.02 mg/g). The extracts showed antioxidant activity by sequestration of DPPH and ABTS radicals. In addition, five phenolic components were identified in the buriti peel and pulp extracts in SUPRAS and the buriti peel ethanolic extract. These extracts showed excellent activity in inhibiting bacterial growth of Staphylococcus aureus (ATCC 29213) and Enterococcus faecalis (ATCC 4028). The solvents were biocompatible with the vegetable matrix, presenting satisfactory yields of beta-carotene and polyphenols. Thus, the present bioactives have shown desirable biological activity for application in cosmetic products.
